High-performance liquid chromatography systems for the analysis of analgesic and non-steroidal anti-inflammatory drugs in forensic toxicology

Abstract

High-performance liquid chromatography retention date are presented for over 40 analgesic drugs on an ODS-silica packing material to assist in the identification of these compounds. Three isocratic eluents prepared from isopropanol, formic acid and an aqueous phosphate buffer have been used. One eluent has been used for the analysis of paracetamol in whole blood.

    HPLC procedures are most often used for the analysis of paracetamol. Common C18 or C8 packing materials for LC columns have been most widely used and can be recommended for separation of paracetamol [4,10,11,19,23,36,59,93–108]. Isocratic or gradient elution procedures using acetonitrile or methanol/buffered water mixtures or mixtures of solvents (acetonitrile, hexane, isopropanol) with phosphate or acetate buffers resulted in sufficient separation.

    This drug produces a relatively high allergy and chemical intolerance [1]. Dipyrone has been determined by titrimetric [2], UV-spectrophotometric [3–5], colorometric [6], and polarographic [7] methods, high performance thin layer chromatography (HPTLC) [5], gas–liquid chromatography (GLC) [8] and liquid chromatography (LC) [9]. Reversed phase LC is the seperation method of choice for most pharmaceutial compounds both hydrophilic and hydrophobic, due to the stable and reproducible nature of the LC columns, the largely aqueous composition of mobile phases and relative ease of reproducing the methods in different laboratories.
