The role of NMDA receptors in the pathophysiology and treatment of mood disorders
2014, Neuroscience and Biobehavioral ReviewsCitation Excerpt :Multiple studies have suggested the positive effects of Mg2+ in the treatment of MDD. In the physiological condition, Mg2+ blocks NMDAR channels through binding with fairly high affinity to a site inside the electrostatic field within the pore (Fig. 1; Reynolds, 1990; Sharma and Stevens, 1996). This prevents other ions (i.e. Ca2+) from passing freely through the channel (Sharma and Stevens, 1996).
Spontaneous synchronized calcium oscillations in neocortical neurons in the presence of physiological [Mg2+]: Involvement of AMPA/kainate and metabotropic glutamate receptors
2004, Brain ResearchCitation Excerpt :Ca2+ spikes also promote neurotransmitter receptor expression and channel maturation in Xenopus embryonic neurons [13] and play a critical role in regulating the advancement of the leading process of migrating cerebellar granule neurons [21]. Synchronized Ca2+ oscillations in cultured neurons were first reported in hippocampal neurons [29], and similar oscillatory behavior has been reported in other neuronal cell types including neocortical neurons [30,35] and cerebellar granule neurons [22,28]; however, these oscillations were typically observed upon removal or lowering of extracellular Mg2+. Mg2+ is a critical ion in the regulation of NMDA receptors [30].
Reverse microdialysis of N-methyl-D-aspartic acid into the lateral hypothalamus of rats: Effects on feeding and other behaviors
2001, Brain ResearchCitation Excerpt :However, the strong correlation we observed between responses suggests that the lower concentrations we employed were not cytotoxic to the LH neurons mediating the elicited feeding. It may also be noteworthy that NMDA receptors can exhibit various forms of plasticity and their activity is modulated by a variety of factors [42,65]. Interestingly, the elicited eating responses in the current experiment remained remarkably stable, arguing that the sensitivity of NMDA receptors and their downstream components were maintained despite the intense stimulation of the receptor.
Zinc in the retina
2001, Progress in NeurobiologyCitation Excerpt :NMDA receptors are structurally complex with separate binding sites for glutamate, glycine, phencyclidine, magnesium, zinc, cadmium, protons and polyamines (Johnson and Ascher, 1987; Kleckner and Dingledine, 1988; McBain and Mayer, 1994; Bigge, 1999) (Fig. 7). The normal function of NMDA receptors depends on a dynamic equilibrium among multiple facilitative and inhibitory factors (Mayer and Westbrook, 1987; Mayer et al., 1992; Aizenman et al., 1989; Levy et al., 1990; Reynolds, 1990; Gozlan and Ben-Ari, 1995). Thus, for example, opening of the NMDA receptor ion channel requires the activation by glutamate, glycine and polyamines and it needs to be phosphorylated.
